Det. Matthews:
John - John picked him, for whatever reason, to be the main test in Saw II. He abducted his son and placed him in the house trap and taunted Eric with needing to know where his son Daniel was. In order to find him, John tested Eric's patience and his ability to follow the rules. All he had to do was sit and listen to him for two hours. John, as he always does, revealed where Daniel was before their game even started, "he's in a safe place." Unable to follow the rules, Det. Matthews brutally beats John, similar to his assault on Amanda, and John tricks him into thinking he's taking him to find his son. What he really did was place him into Amanda's trap where she captured him and leaves him in the bathroom. John also uses Matthews as bait for Amanda in her final tests of willingness to keep people alive and letting them make their own choices. It is still unknown why John picked Det. Matthews.

Det. Tapp - Tapp and Matthews were in the same crime division that followed the Jigsaw murders. Any other direct connection unknown.
Det. Sing - Sing and Matthews were in the same crime division that followed the Jigsaw murders. Any other direct connection unknown.
Det. Kerry - Kerry and Matthews were once partners, as stated in Saw II. It is also safe to assume that they had a love affair resulting in Eric's marriage falling apart as stated by Kerry, "you already lost your son once because you got involved with me." In Saw II the two detectives seem to have a lot of personal tension and anger toward each other. Kerry is the most upset by Eric's disappearance after the events of Saw II. Any further connection is unknown.
Amanda - Matthews framed Amanda for drug possession charges and sent her to prison under false accusations. It is unknown how or why she was framed by him. He is locked in the bathroom after the events of Saw II and brutally beats her after finding her in the hallways in Saw III. He was originally killed by Amanda in Saw III but it was changed so he was saved and held captive until Rigg's test.

Sgt. Rigg - Was an old colleague of Matthews and worked in the same division that followed the Jigsaw murders. He was ultimately killed by Rigg after he neglected to follow the rules of the Cherish Your Life test.
Daniel - Daniel is Eric's son.
Michael - He was Matthew's informant. It was at the scene of Michael's trap that Eric finds the hint that Jigsaw is looking for him.
Gus - According to John, Eric planted evidence to obtain a conviction and was Gus' arresting officer. It is unknown what evidence was planted and why. Any other direct connection is unknown.
Xavier - According to John, Eric planted evidence to obtain a conviction and was Xavier's arresting officer. It is unknown what evidence was planted and why. Any other direct connection is unknown.
Addison - According to John, Eric planted evidence to obtain a conviction and was Addison's arresting officer. It is unknown what evidence was planted and why. She mentions after seeing a picture of Daniel and Eric in the house that he set her up. Director comments backed up the theory and said Eric went to her as a customer and busted her. Hoffman (mind you I haven't watched Saw V in a while so I'll do my best):
John - He was recuited by John as an apprentice after he tried to stage a murder of his own to be a Jigsaw murder. Seeing that I explained it already, you get the basics.
Paul - Aided John in capturing Paul in his car. Any other direct connection is unknown.
Mark - Watched through the hole in the wall during the trap. Any other direct connection is unknown.
Zep - Direct connection is unknown.
Paul - Direct connection is unknown.
Det. Tapp - Worked in the same police precinct. Any other direct connection is unknown.
Det. Sing - Worked in the same police precinct. Any other direct connection is unknown.
Det. Kerry - Was on the scene after Troy's game was carried out alongside Kerry and Rigg. It is suggested by Strahm that someone else helped in Kerry's angel trap. Anyone with a brain can see that person is Hoffman.
Amanda - The dynamics of their relationship is still fairly unknown. We know that he wrote her the upsetting letter in Saw III and that he constantly tried to change John's mind about Amanda, examples being while setting up the Saw II house and just before she brings Lynn into the sick room. Hopefully their relationship will be explained more in Saw VI.

It is safe to assume that Hoffman is the main planner for the Cherish Your Life test and created it the way he did knowing what kind of a person Rigg was. He warned him about not going through uncleared doors. Hoffman also was the main reason Rigg left the police station the night they found Kerry. He told him he was overworked and had to go home.

Jill - Was seen questioning her in Saw IV early on in the Jigsaw murders. It is unknown whether they knew of and/or worked with/against each other (in terms of something other than police questioning). This is another relationship that might be expanded in Saw VI.
Agent Strahm - Strahm quickly grew to be an enemy of Hoffman. When surveying Kerry's murder scene Strahm's questions and answers, that were correct, made Hoffman uneasy. We see in Saw V that he set up Strahm to be captured and killed in the Water Cube trap. It was all part of the design to leave Hoffman the last one standing from the Gideon building. Strahm unexpectedly survives and Hoffman lures him into yet another trap knowing Strahm had started to figure out his involvement with Jigsaw. Hoffman locks himself into the glass box and watches Strahm die. A similar unwinnable theme which suggests Amanda was set up the entire time. Any other direct connection is unknown.
Agent Perez - He also came in contact with her at Kerry's murder scene. He had a brief conversation with Perez about the children's clothing he was carrying (obviously it was for Corbett, Lynn and Jeff's daughter) and quickly dismissed Perez's comments to it. Any other direct connection is unknown.
Art - Was in the Cherish Your Life trap alongside Art and Eric. Hoffman was kept alive by Art during the trap because Eric kept jumping off of the ice block and Art would place him back on. Any other direct connection is unknown. Art was also the lawyer on scene when Rigg punched Rex in the school. Hoffman covered up Rigg's mistake and Art could tell he was lying.
Cecil - Direct connection is unknown.
Gideon - Set up some of the traps in the Gideon building, including the one he was held in. He set up everything to make him the sole survivor. Any other direct connection is unknown.
Ivan - Direct connection is unknown.
Brenda - Direct connection is unknown.
Morgan - Was on scene when Rigg punched out Rex at the school after questioning a child abuse case. Any other direct connection is unknown.
Rex - Was on scene when Rigg punched him out in the school. Any other direct connection is unknown.
Dr. Heffner - Called Hoffman to John's autopsy after the "play me" tape was found inside of his stomach. Any other direct connection is unknown.

Agent Erickson - Hoffman became tied with Erickson when Strahm began poking around old police files trying to place Hoffman at the scene of each of the victims. Hoffman shared conversations about Strahm's state of mind with Erickson and placed him at the scene of the Final Five. The result of that is still unknown and should be revealed in Saw VI.
The Fatal Five - Placed all of the victims in the trap. Any other direct connection is unknown at this point.
Seth - Seth murdered Hoffman's sister. Hoffman captured him, created the pendulum trap to mock a Jigsaw murder, and killed him. Needless to say he got away with it. The only one to notice was John. Hoffman's actions with Seth led to his recruitment.
And now my mind is blown. I agree with my buddy, though. Guaranteed most, if not all, of these characters will get tied together somehow.
